Overview

FT538 + daratumumab is a **combination therapy** comprising FT538, an off-the-shelf induced pluripotent stem cell (iPSC)-derived, multi-engineered natural killer (NK) cell product, paired with daratumumab, a CD38-targeting monoclonal antibody. FT538 is genetically modified to lack CD38 (to avoid daratumumab-induced NK cell fratricide), expresses a high-affinity non-cleavable CD16 Fc receptor for enhanced antibody-dependent cellular cytotoxicity (ADCC), and an IL-15/IL-15 receptor fusion protein for improved persistence. Daratumumab specifically targets CD38 on malignant myeloma cells, inducing cell death through ADCC, complement-dependent cytotoxicity (CDC), and antibody-dependent cellular phagocytosis (ADCP). Together, this combination is designed to maximize tumor cell killing in diseases like multiple myeloma and potentially acute myeloid leukemia, by enabling daratumumab-driven NK cell cytotoxicity while preserving NK cell survival, persistence, and function[1][3][4][7].
