Overview

FT538 + elotuzumab is an investigational combination therapy composed of FT538, a multiplex-engineered, allogeneic natural killer (NK) cell therapy derived from a clonal induced pluripotent stem cell (iPSC) line, and elotuzumab, a SLAMF7-directed monoclonal antibody approved for multiple myeloma. FT538 is engineered with three modifications: a high‑affinity, non-cleavable CD16 Fc receptor for enhanced antibody-dependent cellular cytotoxicity (ADCC), an interleukin-15/IL-15 receptor fusion for cytokine-autonomous NK cell persistence, and a CD38 knockout to confer resistance to fratricide induced by anti-CD38 monoclonal antibodies. In combination, elotuzumab mediates targeting and opsonization of SLAMF7-expressing multiple myeloma cells, while FT538 provides cytotoxic function and persistence. The combination is being evaluated for relapsed/refractory multiple myeloma in early-phase clinical studies[1][3][5][9].
