Overview

FT576 is an off-the-shelf, multiplexed-engineered chimeric antigen receptor (CAR) natural killer (NK) cell therapy derived from induced pluripotent stem cells (iPSCs). It is designed for the treatment of multiple myeloma and incorporates four key modifications: - A proprietary CAR targeting B-cell maturation antigen (BCMA) - A high-affinity 158V, non-cleavable CD16 Fc receptor to enhance antibody-dependent cellular cytotoxicity (ADCC) - An IL-15 receptor fusion to promote NK cell activity and persistence - Knockout of CD38 expression to prevent self-fratricide and enable combination with anti-CD38 monoclonal antibodies such as daratumumab These features are intended to provide multi-antigen targeting of myeloma cells, augment ADCC, improve persistence of the therapeutic cells in vivo, and prevent fratricide when used in combination with anti-CD38 therapies. The drug is being developed primarily for relapsed or refractory multiple myeloma[1][3][4][5][6].
