Overview

FT596 is an investigational, universal, off-the-shelf natural killer (NK) cell cancer immunotherapy derived from a clonal master induced pluripotent stem cell (iPSC) line. It is engineered with three active anti-tumor components: a chimeric antigen receptor (CAR) targeting CD19; a high-affinity non-cleavable CD16 Fc receptor to enhance antibody-dependent cellular cytotoxicity and enable combination with monoclonal antibodies such as rituximab (targeting CD20); and an IL-15/IL-15 receptor fusion to promote NK cell survival and proliferation without systemic cytokine support. This multi-antigen targeting approach aims to overcome resistance due to antigen escape in B-cell malignancies. FT596 is designed for scalable, standardized manufacturing and off-the-shelf use in patients with relapsed or refractory B-cell lymphoma and leukemia[1][3][5][6][8].
