Overview

FT596 + obinutuzumab is an investigational combination therapy consisting of FT596, an off-the-shelf, induced pluripotent stem cell (iPSC)-derived chimeric antigen receptor natural killer (CAR NK) cell product targeting CD19, and obinutuzumab, an anti-CD20 humanized monoclonal antibody. FT596 is engineered to uniformly express an anti-CD19 CAR, a high-affinity, non-cleavable CD16 (FcγRIIIa) to enhance antibody-dependent cellular cytotoxicity (ADCC), and a fusion protein of IL-15/IL-15 receptor alpha for persistence and expansion. Obinutuzumab binds CD20 on B cells, enhancing direct and immune-mediated cytotoxicity, especially via NK cell activation and ADCC. The combination is under investigation for relapsed/refractory B-cell lymphoma and chronic lymphocytic leukemia, aiming to increase anti-tumor activity through dual targeting of B-cell antigens and enhancing NK cell-mediated immune responses[1][2][3].
