FT596 + rituximab is a combination cellular immunotherapy consisting of *FT596*, an induced pluripotent stem cell (iPSC)-derived chimeric antigen receptor (CAR) natural killer (NK) cell therapy, administered alongside the monoclonal antibody *rituximab*. FT596 is engineered to express a CAR targeting CD19, a high-affinity, non-cleavable CD16 Fc receptor (to enhance antibody-dependent cellular cytotoxicity [ADCC]), and an IL-15/interleukin-15 receptor alpha fusion to support NK cell persistence and expansion. Rituximab targets CD20 on B cells, facilitating NK cell-mediated cytotoxicity. The combination is under clinical investigation for relapsed or refractory B-cell lymphoma, aiming to leverage multi-antigen targeting (CD19 via FT596, CD20 via rituximab) and improved immune cell persistence and tumor clearance[1][2][3].
