Overview

FT825 + cetuximab is a combination therapy comprised of FT825, an induced pluripotent stem cell (iPSC)-derived chimeric antigen receptor (CAR) T-cell product, and cetuximab, a monoclonal antibody targeting the epidermal growth factor receptor (EGFR). FT825 features a CAR directed at human epidermal growth factor receptor 2 (HER2) incorporating a cancer-selective antigen binding domain (H2 CasMab-2) engineered to minimize off-tumor toxicity. The combination is being studied in advanced solid tumors, particularly HER2-positive cancers, to exploit potential synergistic anti-tumor effects by targeting both HER2 (via FT825) and EGFR (via cetuximab), aiming to enhance tumor clearance and reduce tumor escape. Cetuximab is FDA-approved for other indications as an anti-EGFR therapy. The combination is under clinical evaluation in a Phase 1 study for safety, tolerability, pharmacokinetics, and antitumor activity in patients with advanced solid tumors[2][3][4][5][7].
