Drug intelligence / Profile preview
FT836 + daratumumab is an investigational combination regimen being developed for the treatment of relapsed and/or refractory multiple myeloma. The regimen combines FT836, an off-the-shelf, allogeneic chimeric antigen receptor (CAR) T-cell therapy derived from a multiplexed-engineered induced pluripotent stem cell (iPSC) master cell bank, with daratumumab, an FDA-approved anti-CD38 monoclonal antibody. FT836 is engineered to target the conserved α3 domain of the stress-induced ligands MICA and MICB, which are highly expressed on multiple myeloma cells, thereby preventing proteolytic shedding and tumor escape. Additionally, FT836 incorporates a high-affinity, non-cleavable CD16a receptor (hnCD16) to enable antibody-dependent cellular cytotoxicity (ADCC) when combined with therapeutic antibodies like daratumumab, and features 'Sword & Shield' technology (alloimmune defense receptor and CD58 knockout) to evade host immune rejection without the need for conditioning chemotherapy. The combination is being evaluated in a Phase 1 investigator-initiated clinical trial.
