Overview

FucBB04 is a very high-molecular weight (3700 kDa) fucoidan derived from the brown algae *Laminaria hyperborea.* It is a sulfated polysaccharide investigated for its bioactivity in age-related macular degeneration and other models of oxidative stress and inflammation. Mechanistically, FucBB04 exhibits anti-inflammatory effects by modulating the secretion of cytokines—specifically reducing IL-6 and IL-8 in response to TLR-3 and TLR-4 pathway activation. It also significantly suppresses VEGF secretion in retinal pigment epithelial (RPE) cells after 7 and 28 days of exposure, suggesting antiangiogenic potential. However, unlike other fucoidans of lower molecular weight, FucBB04 lacks short-term antioxidative effects and may negatively impact RPE cellular function, including phagocytosis and RPE65 expression. The extract showed some protective bioactivities but is not recommended for further development for AMD therapeutics due to these negative influences on RPE function[1][3][5][7][11].
