Overview

Fulmetibant is a small molecule bradykinin 1 receptor (BDKRB1) antagonist developed for the treatment of diabetic neuropathic pain. It was evaluated in the BRADiNP multicenter, randomized, placebo-controlled phase II clinical trial for painful diabetic peripheral neuropathy. The mechanism of action involves selective antagonism of the bradykinin 1 receptor, which is implicated in neurogenic inflammation and pain signaling pathways. The clinical trial found that fulmetibant did not provide a meaningful benefit over placebo in reducing pain scores over four weeks of treatment. Adverse events were mostly mild to moderate in severity[2].
