Drug intelligence / Profile preview
Fumagillin (ZGN-201) is a naturally occurring meroterpenoid antibiotic isolated from the fungus *Aspergillus fumigatus*. It acts as a potent, covalent, and irreversible inhibitor of methionine aminopeptidase 2 (MetAP2). Originally developed as an amebicide and antimicrobial agent, fumagillin is approved in France under the brand name Flisint for the treatment of severe diarrhea caused by *Enterocytozoon bieneusi* microsporidial infections in immunocompromised patients. It is also widely used in apiculture to treat Nosema infections in honey bees. In the early 2010s, Zafgen investigated ZGN-201 as a prototype MetAP2 inhibitor for the treatment of obesity and metabolic disorders, demonstrating significant weight loss and fat metabolism regulation in preclinical models, though its direct development for obesity was discontinued in favor of other synthetic analogs.
