Fumaric acid esters (FAEs) are a class of oral small molecule systemic therapies used for the treatment of moderate to severe plaque psoriasis and psoriatic arthritis. They exert their therapeutic effects through immunomodulatory and anti-inflammatory mechanisms, primarily by activating the nuclear factor erythroid 2-related factor 2 (Nrf2) pathway and inhibiting the nuclear factor kappa B (NF-κB) pathway. In Switzerland, the Swiss Dermatology Network for Targeted Therapies (SDNTT) maintains a long-term observational registry to monitor the real-world safety, effectiveness, and quality of life outcomes for patients treated with FAEs and other systemic therapies over a 10-year period. While SDNTT sponsors the registry, the drugs themselves were developed by pharmaceutical companies such as Biogen and Almirall.
