Fumaven is a small molecule therapeutic consisting of fumaric acid esters (FAE), developed by Meroven for the treatment of inflammatory skin conditions, specifically psoriasis and rosacea. Fumaric acid esters exert their therapeutic effects through complex immunomodulatory and anti-inflammatory pathways. These include the inhibition of the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) signaling pathway and the activation of the nuclear factor erythroid 2-related factor 2 (Nrf2) antioxidant response pathway. These actions lead to a shift in cytokine production from a pro-inflammatory Th1/Th17 profile to an anti-inflammatory Th2 profile. Fumaven is currently approved for the treatment of psoriasis and rosacea, with ongoing post-marketing clinical activities.
