Fuzituximab (IMAB001) is a chimeric IgG1 monoclonal antibody that specifically targets Claudin 18.2 (CLDN18.2), a tight junction protein frequently overexpressed in various solid tumors, particularly gastric and pancreatic cancers. Developed by Ganymed Pharmaceuticals (later acquired by Astellas Pharma), fuzituximab was designed to induce tumor cell death through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). While it served as a proof-of-concept for targeting CLDN18.2, it was largely superseded in clinical development by zolbetuximab (IMAB362), which demonstrated superior pharmacological properties and clinical efficacy.
