Drug intelligence / Profile preview
Fuzuloparib + abiraterone + prednisone is an investigational combination regimen being developed by Jiangsu Hengrui Pharmaceuticals for the treatment of prostate cancer, specifically metastatic castration-resistant prostate cancer (mCRPC). The regimen combines fuzuloparib (formerly known as fluzoparib), an orally active small-molecule inhibitor of poly(ADP-ribose) polymerase 1 and 2 (PARP1/2), with abiraterone acetate, a CYP17A1 inhibitor that blocks androgen biosynthesis, and prednisone, a corticosteroid co-administered to manage mineralocorticoid excess. By simultaneously targeting the androgen receptor signaling pathway and DNA damage repair pathways, this combination aims to provide a synergistic therapeutic effect. In the Phase III FUZUPRO trial, the combination demonstrated a significant improvement in radiographic progression-free survival (rPFS) compared to abiraterone and prednisone alone as a first-line treatment for patients with mCRPC, particularly in those with homologous recombination repair (HRR) gene mutations or DNA damage repair deficiencies (DRD).
