Overview

FXS4640 (formerly known as XS-03) is an orally administered, small molecule inhibitor of polo-like kinase 1 (PLK1) being developed by Fosun Pharma and its subsidiary Jiangsu Xingsheng Xinhui Pharmaceutical, in collaboration with NovaOnco JS Therapeutics. PLK1 is a key regulator of mitosis, and its inhibition leads to cell cycle arrest and apoptosis. FXS4640 is designed to exploit synthetic lethality in tumors harboring RAS mutations. It is currently being evaluated in clinical trials for the treatment of advanced solid tumors and RAS-mutated metastatic colorectal cancer (mCRC) in combination with chemotherapy (FOLFOX or FOLFIRI) and bevacizumab.
