FXX489 (also known as NNS309) is a radiopharmaceutical peptide-drug conjugate (RI-PDC) developed by PeptiDream in collaboration with Modulus Discovery. It is designed as a theranostic agent targeting Fibroblast Activation Protein (FAP), which is highly expressed on cancer-associated fibroblasts (CAFs) within the stroma of various solid tumors. The platform utilizes a peptide backbone conjugated to a chelator, allowing for labeling with different radioisotopes: Gallium-68 (68Ga) for PET imaging and diagnostic purposes, and Lutetium-177 (177Lu) for targeted radionuclide therapy. By targeting the tumor microenvironment rather than the cancer cells directly, FXX489 aims to overcome the delivery barriers posed by the dense stroma in tumors such as pancreatic ductal adenocarcinoma (PDAC), non-small cell lung cancer (NSCLC), breast cancer, and colorectal cancer.
