Drug intelligence / Profile preview
Fyarro (nab-sirolimus) is a nanoparticle albumin-bound formulation of sirolimus, a small molecule inhibitor of the mechanistic target of rapamycin (mTOR). Developed by Aadi Bioscience, this formulation utilizes albumin-bound technology to enhance tumor accumulation and target suppression compared to traditional oral mTOR inhibitors. It is the first FDA-approved therapy for adult patients with locally advanced unresectable or metastatic malignant perivascular epithelioid cell tumor (PEComa). The drug specifically targets the mTORC1 complex, which is frequently dysregulated in various cancers, particularly those harboring TSC1 or TSC2 genetic mutations. Fyarro is currently being investigated for its efficacy in other malignant solid tumors with mTOR pathway alterations.
