Overview

Gαq inhibitory peptide, specifically the Gαq-ct variant, is a therapeutic agent designed to disrupt Gαq-dependent signaling pathways. Gαq is a subunit of heterotrimeric G proteins that, when activated by G protein-coupled receptors (GPCRs) such as the endothelin-1 receptor, triggers downstream cascades involving phospholipase C and calcium mobilization. In research conducted by Northwestern University and the University of Chicago, this peptide is being investigated for its potential to treat atrial fibrillation (AF) in the context of heart failure. By blocking Gαq signaling, the peptide reduces arrhythmogenic triggered calcium waves and conduction inhomogeneity associated with atrial fibrosis. The peptide has been evaluated in preclinical models using both cell-permeable formulations (cp-Gαq-ct) and gene therapy approaches involving plasmid delivery via epicardial electroporation.
