G-02-9D7 is a humanized monoclonal antibody clone targeting G-protein-coupled receptor class C group 5 member D (GPRC5D), a cell surface marker highly expressed on malignant plasma cells in multiple myeloma. Developed by KYinno Biotechnology (Beijing) Co., Ltd., the antibody was derived from hybridoma technology using mice immunized with GPRC5D-overexpressing cells. G-02-9D7 exhibits high affinity (EC50 = 1.9 nM) and high specificity for GPRC5D, with no cross-reactivity to related receptors GPRC5A, B, or C. It is primarily utilized as the tumor-targeting arm of a bispecific T-cell engager (BsAb) that also incorporates a CD3-binding domain (clone sp34). This bispecific construct facilitates T-cell redirection and activation, leading to the release of cytokines (IFN-γ, IL-2, IL-8, and TNF-α) and specific cytotoxicity against multiple myeloma cells. Preclinical studies in xenograft mouse models and patient-derived bone marrow samples have demonstrated potent anti-tumor activity, including complete remission of established tumors.
