Overview

G-02-9D7 x sp34 is a humanized bispecific antibody (BsAb) developed by KYinno Biotechnology for the treatment of multiple myeloma. It is designed to simultaneously target G-protein-coupled receptor class C group 5 member D (GPRC5D) on malignant plasma cells and the CD3 epsilon subunit on T cells. By cross-linking these two targets, the antibody facilitates the formation of an immunological synapse, leading to T-cell activation, the release of cytotoxic cytokines (such as IFN-γ, IL-2, IL-8, and TNF-α), and the subsequent redirected lysis of GPRC5D-expressing myeloma cells. Preclinical studies presented at AACR 2022 demonstrated that the G-02-9D7 clone possesses a higher affinity for GPRC5D compared to clinical-stage benchmarks like talquetamab (JNJ-64407564) and shows potent anti-tumor activity in xenograft models, including complete remission of established MM.1S tumors.
