Overview

G-4460 is a phosphorothioate-modified antisense oligonucleotide designed to inhibit the expression of the proto-oncogene protein c-myb. It binds specifically to codon sequences 2 to 9 of c-myb mRNA and blocks translation of this transcript[2][3]. The drug was developed as an antineoplastic agent for hematologic malignancies such as leukemia and chronic myeloid leukemia. Its mechanism involves direct inhibition of c-myb, a transcriptional activator implicated in cell proliferation and differentiation in hematopoietic cells[1][3]. Originally developed by Temple University and the University of Pennsylvania, with later involvement from Genta.
