Overview

G-CSF-primed peripheral blood stem cells (G-PBSCs) are a type of allogeneic or autologous transplant graft used in hematopoietic stem cell transplantation (HSCT). These cells are obtained by administering granulocyte colony-stimulating factor (G-CSF) to a donor, which mobilizes hematopoietic stem and progenitor cells (HSPCs) from the bone marrow into the peripheral blood, where they can be collected via apheresis. G-PBSCs are the most common source of stem cells for adult HSCT due to the ease of collection and rapid hematopoietic recovery (engraftment) compared to bone marrow-derived cells. They are used to treat a wide range of conditions, including acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), myelodysplastic syndromes, and severe aplastic anemia. In the context of haploidentical transplantation, G-CSF priming of the donor is a critical component of protocols like the Beijing Protocol to ensure adequate cell yields and modulate the graft's T-cell composition.
