Drug intelligence / Profile preview
G-TAK is an investigational combination therapy designed to address the dual pathophysiological drivers of acute-on-chronic liver failure (ACLF): excessive systemic inflammation and impaired liver regeneration. It consists of two active components: resatorvid (TAK-242 / YAQ005), a small molecule Toll-like receptor 4 (TLR4) antagonist, and filgrastim (G-CSF), a recombinant granulocyte colony-stimulating factor. Resatorvid selectively and covalently binds to the intracellular domain of TLR4, preventing the recruitment of adaptor proteins and thereby blocking downstream pro-inflammatory cytokine cascades. Filgrastim acts as an agonist at the G-CSF receptor, stimulating the mobilization of progenitor cells and promoting hepatocyte proliferation to support liver regeneration. G-TAK was invented by researchers at University College London, Charité - Universitätsmedizin Berlin, Leipzig University Medical Center, and Leiden University Medical Center under the EU-funded A-TANGO consortium, and is being clinically developed by Yaqrit and its subsidiary Hepyx. It is currently in Phase II clinical trials for the treatment of ACLF and steroid-resistant alcohol-related hepatitis.
