Overview

G00101 is a selective, next-generation small molecule inhibitor of p38 alpha mitogen-activated protein kinase (MAPK14), currently under development by GEn1E Lifesciences. The drug is primarily indicated for the treatment of Acute Respiratory Distress Syndrome (ARDS) and Acute Lung Injury (ALI). By targeting the p38 alpha pathway, G00101 aims to modulate the excessive inflammatory response and cytokine production—including TNF-alpha and IL-6—that leads to pulmonary edema and respiratory failure. G00101 is designed to overcome the toxicity and efficacy limitations observed in first-generation p38 inhibitors, featuring a more favorable safety profile and high potency for oral administration. It was identified and optimized using an AI-driven platform to target inflammatory signaling while minimizing off-target effects.
