Overview

G100 + rituximab is a **combination investigational immunotherapy** composed of G100, a synthetic Toll-like receptor 4 (TLR4) agonist, and rituximab, a chimeric monoclonal antibody targeting the CD20 antigen on B cells. G100 is designed to activate innate and adaptive immune responses within the tumor microenvironment, promoting tumor antigen presentation and T-cell infiltration. Rituximab induces B-cell depletion through antibody-dependent cellular cytotoxicity, complement-mediated cytotoxicity, and direct apoptosis. The combination aims to augment antitumor immunity via both **direct B-cell targeting (CD20)** and **innate immune stimulation (TLR4)**. It is primarily investigated for treatments in relapsed or refractory B-cell non-Hodgkin lymphomas, notably follicular lymphoma and diffuse large B-cell lymphoma. G100 was developed by Immune Design, and rituximab is commercially developed and manufactured by multiple companies[4].
