Overview

G17DT immunogen is a therapeutic cancer vaccine designed to induce an immune response against gastrin 17, a peptide hormone that acts as a growth factor for several gastrointestinal cancers, including pancreatic, gastric (stomach), and colorectal cancers. The drug consists of a synthetic peptide mimicking part of the gastrin 17 hormone conjugated to diphtheria toxoid as a carrier protein. Upon administration, it stimulates the production of antibodies that neutralize both gastrin 17 and its precursor forms, thereby blocking their interaction with the cholecystokinin-2 receptor on tumor cells. This mechanism inhibits tumor growth driven by gastrin signaling. Developed originally by Aphton Corporation and later advanced by Cancer Advances, G17DT has undergone Phase III clinical trials in advanced pancreatic cancer (as monotherapy and in combination with gemcitabine) and Phase II/III trials in gastric cancer (with chemotherapy). It has also been studied for colorectal cancer and gastroesophageal reflux disease (GERD). The product received orphan drug status for pancreatic and gastric cancers in multiple regions[1][2][4][7].
