G207 is a genetically engineered, neuroattenuated, replication-competent recombinant herpes simplex virus type 1 (HSV-1) with oncolytic activity. It has been modified by deleting both copies of the viral genes encoding ICP34.5 and inserting a LacZ gene into the ICP6 gene, which restricts its ability to replicate in normal cells while allowing selective replication in tumor cells. This enables the virus to infect and destroy cancer cells—particularly gliomas and other brain tumors—while sparing healthy tissue. The drug is being developed as an investigational therapy for pediatric and adult brain tumors such as glioblastoma, with demonstrated anti-tumor effects including direct cytopathic action against tumor cells and inhibition of angiogenesis[1][2][3][4][5].
