G3-01

G3-01 (often referred to as G-Cubed) is an orally bioavailable small molecule inhibitor of Galectin-3, a beta-galactoside-binding lectin that plays a pivotal role in the pathogenesis of tissue fibrosis and chronic inflammation. Developed by G3 Pharmaceuticals, the drug is primarily targeted at treating Heart Failure with Preserved Ejection Fraction (HFpEF) and Chronic Kidney Disease (CKD). Galectin-3 is significantly upregulated in response to cardiac and renal injury, where it acts as a mediator of fibroblast activation, collagen deposition, and macrophage-driven inflammation. By binding to the carbohydrate-recognition domain of Galectin-3, G3-01 aims to block these pro-fibrotic signaling pathways, potentially preventing or reversing the structural remodeling of the heart and kidneys and improving clinical outcomes in patients with these progressive conditions.
