G3335 is a small molecule selective antagonist of the Peroxisome Proliferator-Activated Receptor gamma (PPAR-γ). In the context of hematological oncology research, particularly multiple myeloma (MM), G3335 is used to investigate the role of the bone marrow microenvironment in disease progression. Malignant plasma cells in MM often induce the differentiation of mesenchymal stem cells (MSCs) into bone marrow adipocytes (BMAs) via the PPAR-γ signaling pathway. These BMAs, in turn, support myeloma cell growth, migration, and chemoresistance by secreting pro-survival cytokines such as IL-6, SDF-1α, and IGF-1. G3335 acts by blocking PPAR-γ, thereby inhibiting adipogenesis and the subsequent release of these cytokines. This pharmacological intervention has been shown to partially reverse the protective effects of adipocytes on myeloma cells, sensitizing them to chemotherapeutic agents like bortezomib.
