G36-CD28-TCRζ CAR-T cells are an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target Glypican-3 (GPC3), a cell-surface oncofetal antigen highly expressed in hepatocellular carcinoma (HCC) and other solid tumors of embryonic origin. The CAR construct features a humanized single-chain variable fragment (scFv) derived from the G36 antibody for high-affinity antigen recognition, coupled with a CD28 co-stimulatory domain and a TCRζ (CD3ζ) signaling domain. This combination provides the necessary signals for T-cell activation, proliferation, and cytotoxic activity upon binding to GPC3-expressing cancer cells. Primarily developed for the treatment of advanced GPC3-positive liver cancer, this therapy aims to induce targeted tumor cell lysis while minimizing off-target toxicity in normal tissues where GPC3 expression is absent.
