Drug intelligence / Profile preview
G-4544 is a proprietary oral small molecule formulation of gallium nitrate, developed by Genta with delivery technology from Emisphere Technologies. It is being investigated as a treatment for diseases associated with accelerated bone loss, such as osteoporosis, Paget's disease, and bone metastasis. Its active ingredient, gallium nitrate, works by inhibiting osteoclast activity, preventing bone breakdown, and lowering blood calcium levels. It also incorporates into hydroxyapatite, promoting crystallization and reducing bone mineral solubility. The injectable form of gallium nitrate, branded as Ganite, is approved for the treatment of cancer-related hypercalcemia. Additionally, gallium nitrate exhibits antitumor activity, particularly in lymphoma, by binding to transferrin, disrupting iron transport, blocking ribonucleotide reductase, and inducing apoptosis.
