G47ΔmADK-S is an oncolytic herpes simplex virus type 1 (oHSV-1) engineered to express the short isoform of mouse adenosine kinase (mADK-S). It is based on the G47Δ backbone, a third-generation oHSV-1 characterized by deletions in the γ34.5 and ICP47 genes and an inactivation of the ICP6 gene. The virus is designed to target the immunosuppressive adenosine pathway within the tumor microenvironment (TME) of glioblastoma (GBM). By expressing ADK-S, the virus facilitates the phosphorylation of extracellular adenosine into adenosine monophosphate (AMP), thereby reducing adenosine-mediated immunosuppression and potentially enhancing anti-tumor immune responses. It is currently being evaluated in preclinical models for the treatment of glioblastoma.
