G47delta-KillerRed is an experimental photodynamic oncolytic virotherapy designed for the treatment of central nervous system malignancies, including high-grade gliomas and malignant meningioma. It consists of G47delta, a third-generation genetically engineered oncolytic herpes simplex virus type 1 (oHSV-1), which has been modified to express KillerRed, a genetically encoded photosensitizing fluorescent protein. The therapeutic approach combines the direct tumor-lytic effects of the oncolytic virus with photodynamic therapy (PDT). Upon infection of tumor cells, G47delta-KillerRed expresses the KillerRed protein, which, when activated by specific light irradiation (e.g., amber LED or laser), generates reactive oxygen species (ROS) to induce further tumor cell death. This dual-mechanism strategy aims to enhance tumoricidal efficacy and is currently being investigated in preclinical models by researchers at Massachusetts General Hospital.
