G8 CAR T cells are an experimental adoptive cell therapy consisting of T cells engineered to express a chimeric antigen receptor (CAR) derived from a novel B7-H3-specific nanobody (VHH). Developed by researchers at the National Institutes of Health (NIH), these CAR T cells target B7-H3 (CD276), a transmembrane protein overexpressed in various solid tumors and tumor-associated myeloid cells, while showing limited expression in normal tissues. The G8 clone was isolated from a camelid VHH phage display library and has demonstrated potent in vitro cytolytic activity against B7-H3-positive pancreatic ductal adenocarcinoma (PDAC) and neuroblastoma (NB) cell lines. While other clones like C4 and B12 showed superior in vivo efficacy in metastatic PDAC and NB mouse models in the same study, G8 remains a specific candidate within this nanobody-based CAR-T platform for treating solid tumors.
