GA31-LPC is a macrophage-targeted lipopeptide complex designed to act as a ligand-independent inhibitor of the Triggering Receptor Expressed on Myeloid cells 1 (TREM-1). Developed by SignaBlok, this agent specifically targets TREM-1 signaling on macrophages within the tumor microenvironment (TME), distinguishing it from broad TREM-1 inhibitors that affect all TREM-1-expressing myeloid cells. In preclinical models of pancreatic cancer, GA31-LPC has demonstrated the ability to inhibit tumor progression and overcome resistance to PD-L1 checkpoint blockade when used in combination therapy. Its therapeutic efficacy is highly dependent on the timing of administration relative to other treatments, showing superior outcomes when administered after chemotherapy in certain xenograft models.
