Gabapentin + oxaliplatin refers to the co-administration of gabapentin, an anticonvulsant and neuropathic pain agent, with oxaliplatin, a platinum-based chemotherapy drug. Oxaliplatin is primarily used in the treatment of colorectal cancer and works by forming DNA crosslinks that inhibit DNA replication and transcription, leading to cell death. Gabapentin acts on voltage-gated calcium channels (specifically the alpha-2-delta subunit) in the central nervous system to reduce neuronal excitability and is commonly used for neuropathic pain. The combination has been studied for its potential to reduce or prevent oxaliplatin-induced peripheral neuropathy—a common dose-limiting side effect of oxaliplatin therapy. However, clinical studies have not demonstrated significant benefit from adding gabapentin to standard regimens in reducing the incidence or severity of neurotoxicity caused by oxaliplatin[1][4][6].
