GAd-PEV is a personalized cancer vaccine vector based on a nonhuman Great Ape Adenoviral (GAd) platform. It is designed to encode patient-specific tumor neoantigens identified through next-generation sequencing of an individual’s tumor biopsy. The vaccine is manufactured individually for each patient and administered as the priming component in a prime/boost regimen, with MVA-PEV (a modified Vaccinia Ankara vector encoding the same neoantigens) used as the booster. GAd-PEV aims to elicit robust T-cell responses against cancer cells by presenting these unique neoantigens, thereby enhancing anti-tumor immunity. It has been studied in combination with pembrolizumab, an anti-PD-1 checkpoint inhibitor, primarily in patients with metastatic melanoma and non-small cell lung cancer[1][2][3][4][6].
