GAd20-209-FSP is a non-replicating great ape adenoviral vector vaccine encoding tumor-specific neoantigens (frameshift peptides, FSPs) that are shared across microsatellite instability (MSI) tumors. It is used as the priming component in a heterologous prime/boost genetic cancer vaccine regimen known as NOUS 209. The booster component is MVA-209-FSP, based on Modified Vaccinia Ankara virus. This vaccine aims to induce robust T cell responses against recurrent frameshift neoantigens found in both sporadic and hereditary MSI tumors, including those associated with Lynch syndrome and mismatch repair deficient (dMMR)/MSI-high colorectal cancer. The mechanism of action involves immunostimulation by presenting multiple shared tumor-specific neoantigens to the immune system to elicit anti-tumor immunity[2][4][6][7][9].
