Overview

GADLEN (CD19) is a preclinical-stage bi-functional fusion protein developed by Shattuck Labs. It is part of the GADLEN (Gamma Delta T Cell Engager) platform, which is designed to activate and direct Vγ9Vδ2 T cells toward tumor cells. The molecule consists of the extracellular domains of butyrophilin 2A1 (BTN2A1) and butyrophilin 3A1 (BTN3A1) fused to a single-chain variable fragment (scFv) targeting CD19. This configuration mimics the natural BTN2A1/3A1 heterodimer required for γδ T-cell receptor activation, thereby inducing T-cell proliferation, cytokine release (IFNγ, TNFα), and potent cytotoxicity against CD19-expressing B-cell malignancies. Beyond oncology, the drug is also being investigated for its potential to deplete B cells in autoimmune conditions.
