Gadolinium-conjugated liposomes are a diagnostic and drug-delivery platform consisting of lipid nanoparticles (liposomes) surface-modified or loaded with gadolinium chelates. These liposomes are designed to serve as MRI-trackable carriers for therapeutic payloads, such as RNA interference (RNAi) or CRISPR/Cas9 components. In clinical research, particularly for glioblastoma multiforme (GBM), they are used in conjunction with Magnetic Resonance-guided Focused Ultrasound (MRgLIFUS) to facilitate and monitor the systemic delivery of drugs across the blood-brain barrier (BBB). The gadolinium component allows for the quantification of liposome accumulation in the brain parenchyma using MRI sequences (e.g., ΔR2 mapping), providing real-time feedback on the efficacy of the delivery method.
