Overview

Gadolinium lipid nanoemulsion (Gd-nanoLE) is a research-stage nanoparticle drug delivery system developed by academic researchers at Kobe Gakuin University for use in gadolinium neutron-capture therapy (GdNCT). The formulation consists of gadolinium-diethylenetriaminepentaacetic acid-distearylamide (Gd-DTPA-SA) incorporated into lipid nanoemulsion particles, typically featuring a soybean oil core and a surface layer of hydrogenated phosphatidylcholine and a cosurfactant like HCO-60. These particles are engineered to be smaller than 100 nm to enhance tumor accumulation through the enhanced permeability and retention (EPR) effect. Once localized in tumor tissue, the gadolinium serves as a capture agent for thermal neutrons; the resulting nuclear reaction releases high-energy radiation that selectively destroys cancer cells. Preclinical studies in melanoma-bearing hamsters have demonstrated significant tumor accumulation and growth suppression potential.
