Gadoquatrane is a low-dose **macrocyclic gadolinium-based contrast agent** developed by Bayer for **contrast-enhanced magnetic resonance imaging**. It is a diagnostic injectable MRI contrast agent rather than a therapeutic drug. Bayer developed gadoquatrane to provide lesion visualization and tissue contrast enhancement at a lower gadolinium exposure than conventional macrocyclic GBCAs; pivotal QUANTI Phase 3 studies evaluated a dose of **0.04 mmol Gd/kg body weight** and showed non-inferior visualization and lesion detection versus approved macrocyclic comparators dosed at **0.1 mmol Gd/kg**. Publicly available Bayer materials identify it as BAY 1747846, and the product was later approved in the United States under the brand name **AMBELVIST** for contrast-enhanced MRI in central nervous system and non-CNS body regions, including pediatric use. Mechanistically, it acts through the **paramagnetic signal-enhancing properties of chelated gadolinium**, shortening proton relaxation times in areas where the agent distributes and thereby improving MRI conspicuity of abnormal vascularity and tissue lesions.
