Overview

Gadoxetate disodium is a gadolinium-based contrast agent (GBCA) used in magnetic resonance imaging (MRI), primarily for the detection and characterization of liver lesions. It is a paramagnetic compound that develops a magnetic moment when placed in a magnetic field, enhancing the relaxation rates of water protons and increasing signal intensity on MRI scans. Gadoxetate disodium exhibits biphasic pharmacokinetics—initially distributing in the extracellular space after intravenous injection and then being selectively taken up by hepatocytes due to its ethoxybenzyl moiety. This selective uptake allows for improved visualization of normal liver tissue versus lesions lacking functional hepatocytes or organic anion-transporting polypeptides (OATPs). The drug is excreted via both renal and biliary pathways. It was developed to improve diagnostic accuracy for hepatic imaging[1][3][4][6][8].
