Overview

GAIA-Aβ is a chimeric fusion protein drug developed as a novel immunotherapeutic agent for Alzheimer's disease. It consists of a single-chain variable fragment of an amyloid-β (Aβ)-targeting monoclonal antibody fused to a truncated receptor-binding domain of growth arrest-specific 6 (Gas6). This construct harnesses TAM (TYRO3, AXL, and MERTK) receptor-dependent phagocytosis to induce clearance of Aβ plaques by both microglia and astrocytes, without triggering Fc receptor-mediated inflammatory responses commonly seen with conventional Aβ antibodies. The dual phagocytic activation and anti-inflammatory mechanism distinguishes GAIA-Aβ from standard antibody drugs, reducing side effects such as cerebral edema and microhemorrhage (ARIA) associated with antibody-mediated clearance in Alzheimer's disease[1][2]. Developed by Illimis Therapeutics, GAIA-Aβ is designed to overcome safety and efficacy limitations of traditional antibody therapeutics in neurodegenerative and immune diseases[2].
