Gal-1 scFv-CD28-4-1BB-CD3ζ CAR-T cells are a third-generation chimeric antigen receptor (CAR) T-cell therapy designed to target Galectin-1 (Gal-1), a carbohydrate-binding protein that is significantly overexpressed in human glioblastoma (GBM), particularly in cases resistant to anti-VEGF therapy. The CAR construct features a codon-optimized single-chain variable fragment (scFv) specific to Gal-1, integrated with the CD3ζ signaling domain and the co-stimulatory cytoplasmic domains of CD28 and 4-1BB. Developed by researchers at the Medical University of South Carolina, these CAR-T cells are engineered from peripheral blood mononuclear cells using a lentiviral vector system. Preclinical studies in vitro and in ex vivo brain slice culture models have demonstrated that these cells selectively kill Gal-1-positive GBM cells and induce the secretion of pro-inflammatory cytokines such as IL-2, IFN-γ, and TNF-α, suggesting potential efficacy for the treatment of glioblastoma.
