Gal-Nab2 is a murine monoclonal antibody (IgG1-kappa) that specifically targets and neutralizes extracellular Galectin-9 (Gal-9). It binds to a linear epitope within the N-terminal carbohydrate recognition domain (N-CRD) of Gal-9, specifically residues 213–224. Galectin-9 is a tandem-repeat galectin that acts as a pleiotropic immune regulator; in the tumor microenvironment, it often promotes immune escape by inducing apoptosis in TIM-3-expressing T cells, while in autoimmune conditions like rheumatoid arthritis, it stimulates the production of pro-inflammatory cytokines such as IL-6 and MCP-1 from fibroblast-like synoviocytes (FLSs). Gal-Nab2 has been shown in preclinical studies to restore T-cell viability and reduce synovial inflammation by blocking these Gal-9-mediated pathways. It is primarily used as a research tool to investigate the therapeutic potential of Gal-9 neutralization in oncology and rheumatology.
