Galectin-1 neutralizing antibodies represent a class of research-stage monoclonal antibodies designed to bind and inhibit Galectin-1 (Gal-1), a highly conserved β-galactoside-binding protein. Gal-1 is frequently upregulated in the tumor microenvironment and by certain hematopoietic cells, where it promotes tumor angiogenesis and mediates immunosuppression by inducing T-cell apoptosis and fostering an environment conducive to tumor growth. Beyond oncology, recent research has implicated Gal-1 as a mediator of thrombotic risk in myeloproliferative neoplasms (MPNs), such as polycythemia vera and myelofibrosis, by acting as a platelet agonist through interactions with αIIbβ3 and GPVI receptors. By neutralizing Gal-1, these agents aim to restore anti-tumor immunity, inhibit pathological vessel formation, and potentially mitigate pro-thrombotic phenotypes in hematologic disorders.
