The galectin-3 binding protein (Gal3BP) mimetic peptide is a therapeutic agent designed to disrupt the interaction between Chitinase-3-like-1 (CHI3L1) and Galectin-3 (Gal3). In glioblastoma (GBM), the CHI3L1-Gal3 protein complex promotes an immunosuppressive tumor microenvironment by driving the migration and infiltration of protumor M2-like macrophages and activating PD-1/PD-L1 signaling. Gal3BP naturally competes with Gal3 for binding to CHI3L1, acting as a negative regulator of this immunosuppressive axis. The mimetic peptide recapitulates this competitive inhibition, leading to decreased M2-like macrophage migration, increased CD8+ T cell infiltration, and reduced expression of PD-L1 and PD-L2 in tumor cells. Developed by researchers at the University of Pittsburgh, this peptide has demonstrated the ability to inhibit tumor progression in both in vitro and in vivo glioblastoma models.
