Overview
Galegenimab is a monoclonal antibody Fab fragment developed as an intravitreal injection for the treatment of geographic atrophy secondary to age-related macular degeneration (AMD). It acts as an inhibitor of high-temperature requirement A serine peptidase 1 (HTRA1), a protein implicated in the progression of dry AMD. Despite robust target engagement, phase 2 clinical trials showed that galegenimab did not significantly slow the progression of geographic atrophy compared to placebo, and development was discontinued due to lack of efficacy and safety concerns related to intraocular inflammation. The drug was originally developed by Genentech, a subsidiary of Roche[1][2][3][4][5].
